The combination works by targeting different stages of the Eimeria life cycle, ensuring comprehensive control:

Nicarbazin: Inhibits the development of Eimeria gametocytes (sexual stage) and schizonts (asexual stage), preventing parasite reproduction and oocyst shedding.

Maduramicin: Disrupts ion transport (sodium, potassium, calcium) across the Eimeria cell membrane, causing osmotic imbalance and death of the parasite’s sporozoite and merozoite stages (early invasive stages).

This dual action not only enhances efficacy against drug-sensitive Eimeria but also reduces the risk of resistance— a major advantage over single-ingredient anticoccidials.

Prohibited Species (Critical!)

The combination premix is strictly forbidden for the following species due to high toxicity or lack of efficacy:

Egg-laying hens (layers): Nicarbazin accumulates in eggs, posing human health risks; maduramicin may also affect egg production.

Breeder chickens: Risk of residue transfer to eggs or chicks; use single-ingredient premixes (if needed) under veterinary supervision.

Ducks, geese, pigeons: Maduramicin is highly toxic to these species (even low doses cause mortality).

Livestock (cattle, sheep, pigs) and companion animals (dogs, cats): No efficacy against their Eimeria strains; maduramicin is lethal to ruminants.

Never exceed maduramicin limits:

Maduramicin has a narrow safety margin—overdosing (even 2x the recommended dose) causes severe toxicity in poultry (signs: ataxia, paralysis, high mortality).

Uniform mixing is mandatory: Inadequate mixing (clumps of premix) leads to uneven dosing—some birds get overdoses (toxicity), others get sub-doses (treatment failure/resistance). Always pre-mix the combination premix with 10–20% of the total feed before adding to the full batch.

Key Safety and Regulatory Precautions

(1) Mandatory Withdrawal Period

The withdrawal period is the time between the last dose and slaughter, ensuring no drug residues in meat. Violations result in food safety penalties:

Broiler chickens: 5–7 days (varies by product label and local rules).

Turkey poults: 7–10 days.Do not slaughter birds before the withdrawal period ends.

(2) Heat Stress Risk

Nicarbazin increases poultry sensitivity to heat stress (temperatures >30°C/86°F). In warm weather:

Provide cooling (fans, misting, shade).

Reduce stocking density.

Monitor for heat stress signs (panting, wing-spreading, lethargy)—discontinue use if stress is severe.

(3) Resistance Mitigation

While the combination delays resistance, improper use still risks it:

Avoid continuous use across production cycles: Rotate with non-cross-resistant anticoccidials (e.g., diclazuril, toltrazuril) every 1–2 cycles.

Do not use for prolonged treatment: Limit treatment to 7 days max; switch to an alternative if coccidiosis persists.

(4) Overdose Management

Signs of overdose (mostly from maduramicin): ataxia (loss of balance), paralysis, convulsions, rapid mortality (especially in young birds).

Immediate action: Stop feeding medicated feed; switch to plain feed.

Provide fresh water (add electrolytes for dehydration).

Consult a veterinarian—no specific antidote exists; supportive care (e.g., vitamin E/selenium supplements) may reduce mortality.
